Leishmania major pteridine reductase 1 belongs to the short chain dehydrogenase family: stereochemical and kinetic evidence.

Pteridine reductase 1 (PTR1) is a novel broad spectrum enzyme of pterin and folate metabolism in the protozoan parasite Leishmania. Overexpression of PTR1 confers methotrexate resistance to these protozoa, arising from the enzyme's ability to reduce dihydrofolate and its relative insensitivity to methotrexate. The kinetic mechanism and stereochemical course for the catalyzed reaction confirm PT...

Species Differences in Alternative Substrate Utilization by the Antibacterial Target Undecaprenyl Pyrophosphate Synthase

Undecaprenyl pyrophosphate synthase (UPPS) is a critical enzyme required for the biosynthesis of polysaccharides essential for bacterial survival. In this report, we have tested the substrate selectivity of UPPS derived from the mammalian symbiont Bacteroides fragilis, the human pathogen Vibrio vulnificus, and the typically benign but opportunistic pathogen Escherichia coli. An anthranilamide-c...

Vibrio harveyi NADPH:FMN oxidoreductase: preparation and characterization of the apoenzyme and monomer-dimer equilibrium.

A rapid chromatography method was developed for the preparation of apoenzyme of Vibrio harveyi NADPH:FMN oxidoreductase with > or =80% yields. The apoenzyme bound one FMN per enzyme monomer with a dissociation constant of 0.2 microM at 23 degrees C. The reconstituted holoenzyme was catalytically as active as the native enzyme. FMN binding resulted in 87 and 92% of quenching of protein and flavi...

Molecular Detection of Vibrio spp. in Fish and Shrimp from the Persian Gulf

Vibrio species are the major seafood-borne bacteria that are frequently associated with the consumption of contaminated sea food. A total of 113 samples including 58 fish and 55 shrimps were studied for the possible contamination with Vibrio species. A biochemical protocol was applied for the identification of the Vibrio isolates and Polymerase Chain Reaction (PCR) was carried out to confirm th...

Molecular Cloning, Overexpression, and Characterization of Steroid-inducible 3a-Hydroxysteroid Dehydrogenase/Carbonyl Reductase from Comamonas testosteroni A NOVEL MEMBER OF THE SHORT-CHAIN DEHYDROGENASE/REDUCTASE SUPERFAMILY*

3a-Hydroxysteroid dehydrogenase/carbonyl reductase (3a-HSD/CR) from Comamonas testosteroni, a bacterium that is able to grow on steroids as the sole carbon source, catalyzes the oxidoreduction at position 3 of a variety of C19–27 steroids and the carbonyl reduction of a variety of nonsteroidal aldehydes and ketones. The gene of this steroid-inducible 3a-HSD/CR was cloned by screening a C. testo...
